IPTG is an effective inducer of protein expression in the concentration range of 100 μM to 1.0 mM. Concentration used depends on the strength of induction required, as well as the genotype of cells or plasmid used. If lacIq, a mutant that over-produces the lac repressor, is present, then a higher concentration of IPTG may be necessary.
In blue-white screen, IPTG is used together with X-gal. Blue-white screen allows colonies that have been transformed with the recombinant plasmid rather than a non-recombinant one to be identified in cloning experiments.
IPTG is a non-metabolizable galactose analog that induces expression of the lac operon in Escherichia coli. IPTG functions by binding to the lac repressor and altering its conformation. This inactivation prevents the repression of theβ-galactosidase coding lac gene. While not a substrate for β-glactosidase, it is a substrate for thiogalactoside transacetylase and has been reported to be an inducer of penicillinase activity in bacteria. IPTG is a commonly used reagent in cloning procedures that require induction ofβ-galactosidase activity and is used in conjunction with X-Gal, in blue-white color selection of recombinant bacterial colonies. Preparation Instructions: IPTG is soluble at 250 mg/ml in water and may be sterilized by filtration through a 0.22 mm filter. The solution may be aliquoted and stored at -20 °C. IPTG solutions can be stored at room temperature for up to one month. When preparing culture plates, aliquots of X-Gal and IPTG may be added to the agar solution after it has been cooled to ~45 °C. Application:IPTG is used in cloning experiments that require the induction of beta-galactosidase activity. It is often used in conjunction with the x-gal or bluo-gal to reorganize the blue-white screening of bacterial colonies that can induce the expression of lac manipulators in e. coli. IPTG plays a role in binding and altering the conformation of lacI repressors to prevent the inhibition of the beta-galactosidase encoding gene lacZ.
